Arbor knot to the spool and a surgeons knot for line to backing.
Don't worry Ma'am....
I'm only here for the Bass.
I use a 12# mono as backing. On most of My reels I'm using a LuckyCraft™ yellow topwater mono, but have ordered a few spools of this stuff for backing purposes:

To attach the lines I use a Uni-to-Uni 4 wraps w/the braid; 5 wraps on the mono side and then moisten both ends prior to cinching.
We fish the Upper Chesapeake Bay for bass, but frequently run into big stripers, blue cats, and even chain pickerel. They can and will run off your main line sometimes, I always use a uni ti uni knot and I always use 14 pound Stren Clear Blue mono for backing. If a big striper hits a big top water bait I want to land it, not cry about what it costs to replace. Quality line is cheap, less than 3 cents a yard, don't skimp. Tie a good quality uni to uni knot and you are good.
I tie backer to spool with an Arbor knot. Backer to main line with a double uni-knot.
I use the end of filler spools for backer. Usually 12# mono or fluorocarbon. Whatever is left in 15 to 20 yard lengths.

I tie 2 improved clinch knots together. I have too little line to fish with on the reel long before I come close to feeling the knot. When I change line I just pull through my fingers until I get to the knot. I boated a 40lb blue cat on 8lb mono with a spinning rod, never came close to the knot.
